En la bioaumentación se emplearon microorganismos eficientes (ME), cultivados en los invernaderos de Lodana. Se abarcaron tres etapas previas: la caracterización del suelo (medio contaminado), la caracterización de los microorganismos eficiente (elemento remediador) y los análisis de aceite quemados (contaminante). Durante los ensayos se determinó el tipo de suelo en Lodana, las especies de microorganismos dentro del consorcio (ME) y los componentes de los aceites quemados (metales pesados, hollín, etc.) Los tres elementos (medio contaminado, contaminante y elemento remediador) se combinaron para el ensayo final: la bioaumentación. Este método se comparó con la atenuación natural, para determinar la eficiencia de cada uno. This research was realized in Lodana, Manabí, Ecuador. It was a study on the efficiency of soil bioremediation contaminated with burned oils, comparing two methods: bioaugmentation and natural attenuation. Efficient microorganisms (EM), cultivated in Lodana's greenhouses, were used for bioaugmentation. Three previous stages were covered: soil characterization (contaminated environment), characterization of efficient microorganisms (remediating element) and burnt oil analysis (contaminant). During the tests, the type of soil in Lodana, the species of microorganisms inside the consortium (EM) and the components of the burned oil (heavy metals, soot, etc.) were determined. In the coming decades, agricultural production will take place in the context of a changing climate, awaiting major changes in the frequency and severity of floods and droughts. Rainfed agriculture will continue to be particularly vulnerable to excesses and deficits in rainfall. This makes it essential to develop sound strategies to limit environmental and economic losses associated with extreme events and regional climate changes. Understanding the links between agroecosystems, water, and food production is important to think about productive strategies by agricultural producers in current and future conditions. El fenómeno de la turbulencia tiene una fuerte presencia en flujos industriales. Sin embargo, el estudio numérico de flujos turbulentos en muchas condiciones de interés está fuera del alcance de la capacidad de cálculo de las supercomputadoras más grandes hasta el momento. La reducción del problema al caso bidimensional permitir obtener soluciones numéricas con parámetros realistas a un menor costo computacional. Cuando se sigue este camino la mayor dificultad reside en la validación experimental de los resultados, ya en que en muchos casos los flujos bidimensionales son de difícil materialización. En esta tesis realizamos un estudio experimental de las propiedades estadísticas de los flujos turbulentos bidimensionales, con especial énfasis en la caracterización de sus leyes de escala y en el desarrollo de fenómenos intermitentes en el flujo. La configuración experimental para flujos bidimensionales considerada consiste en un túnel de jabón donde la turbulencia se genera con un peine. En una primer etapa se plantea el estudio teórico para determinar las condiciones que permiten establecer la analogía entre un flujo en túnel de jabón y un flujo bidimensional incompresible. A continuación tratamos el estudio experimental del campo de velocidades que se realiza con las técnicas de velocimetría PIV y OPFLOW con imágenes obtenidas con las técnicas Schlieren e interferometria. Se estudian las propiedades estadísticas del flujo a partir de 4 diferentes técnicas espectrales: Fourier, Wavelets, Hilbert- Huang y POD Koopman. El estudio ilustra la capacidad de cada una de estas técnicas para recuperar las leyes previstas de manera teórica. El trabajo concluye acerca de la ausencia de intermitencia del flujo estudiado y brinda nuevos elementos de análisis para calificar las bondades de resultados obtenidos con diferentes técnicas numéricas. Turbulence is ubiquitous in atmospheric and oceanic flows. However, the numerical study of threedimensional turbulent flows under realistic conditions and control parameters is out of reach even when using the largest supercomputers up to day. In geophysical flows, the presence of rotation and stratification reduces the dynamics to that of two dimensions, allowing numerical studies to approach the problem with more realistic parameters at a lower computational cost. On the other hand, when this approach is followed the problem resides on experimental validation, since two dimensional flows are hard to obtain in the laboratory. In this proposal we present a pronged study, with experiments to analyze statistical properties of flows in two- and three-dimensions, with a special focus in the characterization of scaling laws and in the development of impulsive events in the flow. The experimental setup to study two-dimensional flows is a soap film “tunnel”, where turbulence will be generated using a comb, while for the three-dimensional case we will use wind tunnels where turbulence is generated by a grid. The velocity field will be measured in the former case using Schlieren velocimetry, while in the latter the PIV and OPFLOW technique will be used. Different spectrals methods will be used to analyze each case. We will study the flow properties in scales larger and smaller than the energy injection scale. As a result of this proposal, we expect to be able to link large scale properties of the flow with spectral properties of turbulence, as well as improve models of hydrodynamic turbulence. Fourier, Wavelet, Hilbert Huang and POD Koopman: statistics flow properties from 4 different spectral techniques are studied. The study illustrates the ability of each of these techniques to recover laws theoretically expected. The thesis concludes about the absence of intermittent flow studied and provides new elements of analysis to describe the benefits of results obtained with different numerical techniques. Este documento se centrará en la contaminación producida por la extracción de diamantes en Sudáfrica, durante la actividad minera y su etapa posterior al cierre desde la perspectiva del Ingeniero Sanitario, se explica de manera general los conceptos generales sobre los diamantes, su origen, yacimientos, métodos de prospección y exploración, los tipos de minería y técnicas mineras para la extracción a cielo abierto y subterráneo, con énfasis principalmente en diamantes provenientes de tuberías de kimberlita como: Kimberley, Koffiefontein, Cullinan y Finsch. Sudáfrica es uno de los principales productores de diamantes en el mundo y además posee todos los tipos de yacimiento existentes, es también un gran caso de estudio porque sus minas son de propiedad privada y están sujetas a regulaciones ambientales lo que permite conocer el manejo por parte de las compañías durante su intervención. La minería de diamantes requiere de un gran movimiento de tierras debido a excavaciones profundas, esto implica el apilamiento de grandes cantidades de material, estos a su vez, expuestos a la intemperie pueden generar lixiviados salinos que pueden ser contaminantes, además las kimberlitas poseen partículas micro finas que permanecen en suspensión y no reaccionan con ningún tipo de floculante por lo que es necesario disponer de presas de lodos donde las partículas se asientan naturalmente y donde los efluentes a tratar son altos en sales, se deben gestionar diversos tipos de residuos minerales y no minerales además de ejecutar planes de rehabilitación y cierre que permitan dar un uso alternativo a las áreas mineras después de la explotación. All types of mining produce an impact on the mining area, sometimes even extending to other populated areas in its surroundings, this can be either destroying the earth's crust, affecting flora and fauna, soil or water contamination, waste generation, atmospheric emissions, etc. This document will focus on the contamination produced by the extraction of diamonds in South Africa, during the mining activity and its post-closure stage from the perspective of the Sanitary Engineer, it will explain the general concepts about diamonds, its origin, deposits, prospecting and exploration methods, types of mining and mining techniques for open pit and underground extraction, with emphasis mainly on diamonds from kimberlite pipes such as: Kimberley, Koffiefontein, Cullinan and Finsch in order to understand how these activities contribute to the impact on the mining areas. South Africa is one of the main diamond producers in the world and has all the existing types of deposits, it is also an exceptional case to study because its mines are privately owned and are subject to environmental regulations, which allows to know the management by the companies during the time of their intervention. Diamond mining requires a large ground movement due to deep excavations; this implies the stacking of large amounts of material that will be later exposed to the elements and can generate polluting saline drainage. In addition, kimberlites have microfine particles that remain in suspension and do not react with any type of flocculant, so it is necessary to have sludge dams where the particles can settle naturally. Asimismo, debido a la robustez de la relación señal/ruido de la técnica, es posible realizar estudios a nivel individual con gran precisión temporoespacial. Es por ello que su aporte a las neurociencias es fundamental. La presente Tesis recurre a esta técnica para combinar métodos clásicos de actividad evocada con el estudio de la conectividad cerebral a gran escala y algoritmos de aprendizaje para estudiar el acoplamiento de redes neuronales en procesos cognitivos clave de la vida cotidiana. Se abordan tres estudios basados en registros invasivos en humanos. En el primero, se investigaron los correlatos cerebrales de una tarea que evalúa el reconocimiento de la intención de daño, un elemento decisivo en la cognición moral. Se encontró que el daño intencional induce actividad temprana (< 200 ms) en la amígdala, con modulaciones que discriminan sistemáticamente entre tipos de situaciones: daño intencional, daño no intencional e interacción no dañina (neutral). Además, los análisis de conectividad mostraron una rápida conexión con regiones prefrontales. Los resultados reflejan la relevancia de la amígdala en la codificación rápida de la intención de dañar, una habilidad crítica para la supervivencia y fundamental en la cognición moral. El segundo estudio se basó en una tarea de reconocimiento de emociones en caras y palabras, dos fenómenos centrales en la interacción social cotidiana. Se analizaron series temporales y métodos de conectividad con el uso de aprendizaje de máquinas estadístico para estudiar cómo el desempeño individual se refleja en la actividad neuronal en una muestra de sujetos con perfiles de desempeño contrastivos. Se mostró que los diferentes perfiles de desempeño se vieron reflejados en su conectividad entre regiones. Este trabajo es una contribución al enfoque de diferencias individuales, una tendencia reciente en el ámbito neurocientífico, que se concentra en estudiar la variabilidad del procesamiento neural a nivel individual. Por último, en el tercer estudio, se utilizaron registros intracraneales y electroencefalográficos para evaluar en qué medida la construcción del sentido semántico está asociada con regiones vinculadas con sistemas multimodales (áreas relacionadas con conceptos abstractos que integran diferentes vías sensoriales) y/o mecanismos corporizados (redes sensoriomotoras específicas para cada modalidad perceptual) involucrados a este proceso. A través de análisis clásicos de oscilaciones junto con aprendizaje automático, se estudió la dinámica de la modulación de estímulos lingüísticos relacionados con partes del rostro (sustantivos como ‘nariz’ y ‘boca’), y la interacción entre los sistemas multimodales y corporizados mediante métodos de conectividad. Se mostró por primera vez que estos estímulos rápidamente modulan marcadores tempranos en regiones de procesamiento de caras (regiones corporizadas), con contribuciones de regiones multimodales, lo cual sugiere que el significado lingüístico depende, en parte, de la reactivación de experiencias sensoriales pertinentes. En su conjunto, los tres estudios de la Tesis brindan aportes directos mediante registros invasivos en humanos para la construcción de modelos de neurociencias cognitivas. Finalmente, los métodos implementados en estos y otros trabajos realizados fueron incluidos en un toolbox propio de análisis de datos. Esta herramienta sirve como un repositorio de métodos con el fin de organizar, reutilizar, compartir y capacitar estudiantes e investigadores de un laboratorio. Si bien está construido con las funciones de los análisis presentados aquí, la principal bondad del toolbox es que es fácilmente extensible y cada laboratorio o grupo de trabajo puede probarlo con sus propios métodos de análisis. Así, la presente Tesis brinda aportes en temáticas relevantes de las neurociencias de registros intracraneales a través de estudios en los que se aplicaron metodologías novedosas, como la conectividad y el aprendizaje automático, contribuyendo además a futuros estudios mediante la creación de una herramienta que facilita y potencia la tarea científica. In recent decades, cognitive neuroscience has shown that cognitive processes rely on dynamic couplings between brain areas over short and long distances. Cognitive events are thus no longer taken to depend solely on a specific region, but rather on the largescale integration of many regions. In human research, most relevant evidence stems from functional magnetic resonance or electromagnetic techniques, marked by limitations in their temporal and spatial resolution, respectively. In this context, invasive intracranial recordings represent a unique opportunity for studying cognition. This technique has a spatial resolution in the order of millimetres and a temporal resolution in the order of milliseconds, alongside one of the best signal-to-noise ratios. Therefore, it is very well suited to evaluating neuroanatomical models and complex temporal dynamics of elemental cognitive domains (e.g., emotion, moral judgement, and language) at the single-subject level. Building on such assets, this thesis relies on intracranial recordings combined with classical evoked activity methods, measures of large-scale brain connectivity, and machine learning approaches to study the coupling of neural networks in key cognitive processes of everyday life. The dissertation comprises three studies. The first one examines neural correlates of the recognition of harmful intentions, a decisive element of moral cognition. Intentional harm induced early amygdalar activity (< 200 ms), with modulations that discriminate among intentional harm, unintentional harm, and neutral actions. Furthermore, connectivity analyses showed a fast coupling with prefrontal regions. These results highlight the key role of the amygdala in the rapid encoding of intentional harm, a critical ability for survival and to moral cognition. The second study assessed emotion recognition in faces and words, two pillars of social interaction. Machine learning algorithms were applied to temporal series data and connectivity coefficients to examine individual signatures of neural activity in subjects with contrastive behavioural performance. The participant’s differential performance profiles were captured by connectivity coefficients. This work represents a contribution to the individual differences approach, a recent neuroscientific trend that focuses on individual neural variability. Lastly, through a combination of intracranial and scalp-level recordings, the third study focused on semantic processing to examine the temporal organization of multimodal systems (areas related to abstract concepts that integrate different sensory pathways) and/or embodied mechanisms (specific sensorimotor networks for each perceptual modality) in the construal of meaning. We studied the dynamics of the modulations of linguistic stimuli denoting to facial body parts (nouns such as ‘nose’ and ‘mouth’) through classical oscillatory analysis together with machine learning, and the interaction between multimodal and embodied systems using connectivity methods. We showed, for the first time, that these stimuli rapidly modulate early markers in facial processing areas (embodied regions), with multimodal contributions, suggesting that linguistic meaning partly relies on fast reactivations of relevant sensory experiences. Together, the three studies provide direct contributions for the construction of models in cognitive neuroscience through invasive recordings in humans. Finally, the methods implemented for these and other works were included in a data analysis toolbox. This tool serves as a repository to ease the organization, reuse, and sharing of scripts, thus supporting student and researcher training. Already equipped with the functions used for present analyses, the toolbox is easily extensible and each laboratory or work group can populate it with their own methods. In sum, building on the uniqueness of intracranial human recordings, the present thesis affords methodological and theoretical contributions in hot topics of cognitive neuroscience, combining novel connectivity and machine learning methods and providing a tool that assists future studies and enhances other potential investigations. Los modelos de microsimulación de tránsito constituyen en la actualidad una herramienta valiosa para el estudio de las arterias urbanas al permitir analizar y visualizar en forma integral la operación de la arteria; esto es, como todos y cada uno de los conductores-vehículos que circulan por la arteria responden a unas dadas condiciones prevalecientes de calzada, tránsito, y control, y características del propio usuario. Permiten ¨ver¨ el funcionamiento del sistema actual o de proyecto, así por ejemplo permiten determinar la formación de colas, proporcionan salidas con parámetros para el análisis y permiten además una animación gráfica, más amigable para el público general. En nuestro medio son utilizados por la Ingeniería de Tránsito sin mayores cuidados en cuanto a su calibración, entre otros, el americano denominado “TRAF-NETSIM” (TRAFFic NETwork SIMulation model), o el europeo “VISSIM” (Verkehr in Städten SIMulation), Traffic in Towns Simulation y el escocés PARAMICS. En esta tesis se pretende encontrar con cierta universalidad y caracterizar estadísticamente el comportamiento de los usuarios en la red arterial de la ciudad de Buenos Aires a través de la medición de dos parámetros: Intervalo de saturación y el tiempo perdido en el arranque, que son cruciales a la hora de modelizar la descarga de una cola de una corriente vehicular. Un objetivo secundario es evaluar la potencialidad, además del método manual tradicional de relevamiento en campo, los métodos de observación y registro en base a videos obtenidos de las cámaras del Sistema Centralizado de Control de Tránsito que posee la Ciudad de Buenos Aires, y por otro lado el análisis de los archivos que entregan los contadores de tránsito, modalidad tipo PVR (per vehicle record). En este tipo de archivo cada paso de un vehículo por el detector constituye un evento a registrar, para lo cual se diseña un protocolo de medición y ensayo piloto respectivo. Los parámetros estudiados permiten en cierto modo el ajuste de los modelos de microsimulación a las condiciones locales, específicamente en lo que hace al comportamiento de los conductores, en el trabajo se utilizarán los resultados para comparar su aplicación a uno de los modelos citados. Los resultados de esta tesis permitirán en primer lugar la calibración de los parámetros aludidos en los programas comerciales de simulación microscópica.
