En esta tesis de Ingeniería Electrónica se propone abordar el tema de Slew Rate en amplificadores de potencia que en la mayoría de los casos resulta ser deficiente induciendo distorsión por intermodulación transitoria (TIM). La ventaja de la topología Current Feedback es la de ofrecer un Slew Rate varias veces superior a las topologías de amplificadores realimentados por tensión, reduciendo la generación de TIM o la distorsión inducida por Slew Rate. El propósito de esta investigación es, además, lograr un amplificador que supere las cifras de distorsión de las topologías básicas de Current Feedback y también de la mayoría de las topologías convencionales, con el objeto de satisfacer al purista más exigente. Para lograr este objetivo, se parte de la topología sencilla original de los primeros amplificadores integrados realimentados por corriente, que no tenían como propósito integrar la cadena de un sistema de audio de alta calidad. La linealidad ofrecida por estos diseños satisfacía ampliamente las características de intermodulación necesarias para un sistema de distribución de TV por cable o amplificador de FI para sistemas de radar y otras aplicaciones en radio frecuencia. Hay muy pocos amplificadores en el mercado con la tecnología de realimentación por corriente con el objeto de tener un alto Slew Rate como premisa de diseño. En su gran mayoría estos diseños replican el formato original de la topologia integrada, de modo que las cifras de distorsión resultan ser poco sorprendentes para los parámetros obtenibles en el presente. Sin embargo, la mayoría de los avances han surgido en el campo de la memoria declarativa, posiblemente debido a la vasta evidencia que sugiere que sólo el aprendizaje dependiente del hipocampo se beneficia del sueño. La reactivación nocturna delas memorias hipocampales parece depender de la sincronía precisa entre las ondas lentas (OL) y los husos del sueño, mecanismo enmarcado en la hipótesis de consolidación de sistemas. En la presente tesis buscamos examinar el rol del sueño en la consolidación de memorias motoras en humanos e identificar los marcadores neurales asociados al proceso de consolidación. Dado que el papel del sueño en la adaptación motora es algo controvertido, en dos experimentos iniciales pusimos a prueba la hipótesis de que el sueño cumple un rol en la consolidación de las memorias de adaptación motora (AM), manipulando la proximidad temporal entre el aprendizaje y el sueño (PAS), de forma que en cada experimento un grupo tuvo la posibilidad de iniciar el sueño ~10 minutos luego de aprender (PAS-10min) y el otro grupo lo hizo ~14 horas después (PAS-14hs). En el Experimento 1, los voluntarios realizaron una sesión de AM a una única perturbación, y en el Experimento 2 realizaron un protocolo de interferencia anterógrada al enfrentarse a dos perturbaciones opuestas separadas por un intervalo de 5 minutos. Encontramos que la proximidad temporal entre el entrenamiento y el sueño aumentó la retención de la memoria en un 30%, un fenómeno que persistió durante al menos dos semanas, lo que sugiere que este tipo de aprendizaje se beneficia del sueño cuando ocurre durante la ventana de consolidación. Sin embargo, no se observó un efecto protector del sueño ante la interferencia. Luego, con el objetivo de identificar los marcadores neurales que apoyan la estabilización dependiente del sueño de las memorias de AM, realizamos otro estudio (Experimento 3) en el que un nuevo grupo de voluntarios fue expuesto a una sesión de AM y a una sesión control (CTL) siguiendo el protocolo PAS-10min. Encontramos que la AM aumentó la densidad global de husos rápidos del sueño (≥12 Hz) pero no lentos (<12Hz) durante el sueño NREM3 (sueño lento profundo). Esta modulación se manifestó localmente, sobre el hemisferio contralateral a la mano entrenada. El aprendizaje motor también aumentó la proporción de husos agrupados en trenes (intervalos inter-huso ≤ 6 segundos), un fenómeno que se ha relacionado recientemente a la reactivación de la memoria. Este efecto se observó únicamente durante NREM3, también de forma local, sobre el hemisferio contralateral a la mano entrenada. Además, esta modulación interhemisférica se correlacionó significativamente con la retención de la memoria de largo término. Para explorar la potencial relevancia del agrupamiento de los husos en la plasticidad dependiente del sueño, cuantificamos el impacto de la AM sobre la duración y amplitud de los husos. Encontramos que ambas características intrínsecas aumentaban en función de la longitud del tren, y alcanzaban valores mayores que los de los husos aislados. También encontramos que la duración y la amplitud de los husos aumentaban en magnitud en función del largo del tren, siendo los últimos husos de un tren más largos y de mayor amplitud que los primeros. Estos hallazgos son consistentes con un rol de los husos en la reactivación de la memoria y la plasticidad neural. Por otra parte, aunque el aprendizaje de adaptación no afectó la densidad de OL, aumentó la potencia de la banda delta (1-4 Hz) localmente, de forma interhemisférica, durante los primeros 30 minutos en comparación con los últimos 30 minutos de sueño NREM, en línea con la hipótesis de homeostasis sináptica (SHY) como un potencial mecanismo que media la estabilización de las memorias durante el sueño. Por último, observamos que la AM aumentó sustancialmente el número de husos rápidos acoplados a la fase activa de las OL (alrededor de su pico positivo), consistente con la hipótesis de consolidación de sistemas. El hecho de que sólo los husos acoplados a una OL (y no los husos desacoplados) predijeron la retención de la memoria luego de dormir, apunta a la relevancia de esta asociación en la consolidación de la memoria motora. Finalmente, para evaluar si la manipulación de la PAS también impacta sobre el acoplamiento huso-OL y sobre la actividad delta, se estudiaron los registros de EEG de ambos grupos del Experimento 1 en el que se observó un beneficio de la proximidad del sueño sobre la retención de la memoria a largo plazo. En el grupo PAS-10min se detectó una modulación en la densidad de husos rápidos durante el sueño NREM (etapas 2 y 3), mostrando un aumento de dicha medida en el hemisferio contralateral a la mano entrenada, respecto al ipsilateral. Además, el acoplamiento entre husos rápidos y ondas lentas también sufrió una modulación interhemisférica. En el mismo sentido, la potencia delta también se moduló de forma interhemisférica al inicio del sueño NREM. Notablemente, estas medidas resultaron moduladas solo en el grupo PAS-10min, en el que el sueño ocurrió dentro de la ventana de consolidación de la memoria de AM, y no en el PAS-14hs. En su conjunto, estos resultados aportan evidencia a favor de mecanismos comunes en la estabilización de las memorias declarativas y motoras. The last decade has seen remarkable advances in the identification of neural markers of sleep-dependent memory consolidation. However, most advances have emerged from the field of declarative memory, possibly because of the vast evidence suggesting that only hippocampal-dependent learning benefits from sleep. Sleep-dependent reactivation of hippocampal memories appears to depend on the precise synchrony between slow oscillations (SO) and sleep spindles, a mechanism framed by the systems consolidation hypothesis. In the present thesis, we sought to examine the role of sleep in the consolidation of motor memories in humans, and to identify neural markers associated with the consolidation process. Given that the role of sleep in motor adaptation is somewhat controversial, in two initial experiments we tested the hypothesis that sleep plays a role in the consolidation of motor adaptation memories (MA) by manipulating the temporal proximity between learning and sleep (PLS), such that in each experiment one group had the opportunity to initiate sleep ~10 minutes after learning (PLS-10min) and the other group did so ~14 hours later (PLS-14hs). In Experiment 1, volunteers performed a MA session to a single perturbation, and in Experiment 2 they performed an anterograde interference protocol by facing two opposite perturbations separated by a 5-min interval. We found that the temporal proximity between training and sleep increased memory retention by 30%, a phenomenon that persisted for at least two weeks, suggesting that this type of learning benefits from sleep when it occurs during the consolidation window. However, a protective effect of sleep against interference was not observed. Next, with the aim of identifying neural markers that support sleep-dependent stabilization of MA memories, we conducted another study (Experiment 3) in which a new group of volunteers was exposed to a MA session and a control session (CTL) following the PLS-10min protocol. We found that MA increased the global density of fast (≥12 Hz) but not slow (<12Hz) sleep spindles during NREM3. This modulation was manifested locally, over the hemisphere contralateral to the trained hand. Motor learning also increased the proportion of spindles clustered in trains (inter-spindle intervals ≤ 6 seconds), a phenomenon that has recently been linked to memory reactivation. This effect was observed only during NREM3, also locally, over the hemisphere contralateral to the trained hand. Moreover, this interhemispheric modulation was significantly correlated with long-term memory retention. To explore the potential relevance of spindle clustering on sleep-dependent plasticity, we quantified the impact of MA on spindle duration and spindle amplitude. We found that both intrinsic features increased as a function of train length, and reached higher values than isolated spindles. We also found that spindle duration and amplitude increased in magnitude as a function of train length, with the latter spindles of a train being longer and of greater amplitude than the former. These findings are consistent with a role for spindles in memory reactivation and neural plasticity. Moreover, although adaptive learning did not affect OL density, it increased delta-band (1-4 Hz) power locally, interhemispherically, during the first 30 min compared to the last 30 min of NREM sleep, in line with the hypothesis of synaptic homeostasis (SHY) as a potential mechanism mediating the stabilization of memories during sleep. Finally, we observed that MA substantially increased the number of fast spindles coupled to the active phase of SO (around their positive peak). The fact that only SO-coupled spindles (and not uncoupled spindles) predicted memory retention after sleep points to the relevance of this association in motor memory consolidation. Finally, to assess whether the PLS manipulation also impacts spindle-SO coupling and delta activity, EEG recordings from both groups of Experiment 1 were studied in which a benefit of sleep proximity on long-term memory retention was observed. In the PLS-10min group, a modulation in the density of fast spindles was detected during NREM sleep (stages 2 and 3), showing an increase in this measure in the hemisphere contralateral to the trained hand, with respect to the ipsilateral one. In addition, the coupling between fast spindles and SO also underwent interhemispheric modulation. In the same vein, delta power was also modulated interhemispherically at the onset of NREM sleep. Notably, these measures were modulated only in the PLS-10min group, in which sleep occurred within the MA memory consolidation window, and not in the PLS-14hs. Taken together, these results provide evidence in favor of common mechanisms in the stabilization of declarative and procedural memories.
