Description An account of the resource Durante el diseño y verificación de estructuras suele ser necesario evaluar la respuesta de las mismas a cargas dinámicas.La determinación analítica o numérica de las frecuencias propias y los modos de vibración es difícil de realizar con precisión,Resulta útil, entonces, el proceso de ingeniería inversa consistente en estimar las propiedades estructurales a partir de la medición de la respuesta.Todas las metodologías desarrolladas con este fin se agrupan bajo el nombre de system identification (identificación de sistemas). En particular, aquellas metodologías que buscan obtener parámetros modales de una estructura se encuadran en lo que se define como experimental modal analysis (análisis modal experimental) o EMA. En la década del 90 se desarrollaron los primeros output-only models, metodologías que trabajan únicamente con la medición de la respuesta.La gran ventaja de estos métodos es que permiten obtener los parámetros modales sin requerir una excitación impuesta, es suficiente con la medición de la vibración ambiente. Es por ello que estos métodos se engloban bajo el término de operational modal analysis (análisis modal operacional) u OMA, midiendo la respuesta de la estructura durante su estado normal de servicio (“operación”) se obtienen los parámetros buscados. Existen dos grandes corrientes respecto a la forma de implementar estos métodos: los que trabajan en el dominio del tiempo, y los que trabajan en el dominio de la frecuencia. En términos generales, los métodos en el dominio del tiempo trabajan ajustando la respuesta a partir de las correlaciones entre señales, mientras que los métodos en el dominio de la frecuencia utilizan las densidades de potencia espectral. Recordando que la relación entre ambas es la Transformada de Fourier, la información que manejan es la misma, pero representada de distinta forma. Esto puede contribuir a que una mayor proporción del cultivo ingrese a la cadena alimentaria, lo que aumenta el valor de la cosecha. De este modo, también es posible limitar la cantidad de arroz quebrado, el cual es usualmente destinado a la alimentación animal, ya que posee un menor valor comercial. Se han ajustado en esta tesis, diversos modelos matemáticos a los datos experimentales, y se han comparado mediante un análisis estadístico meticuloso. Se han elegido de este modo las expresiones que permiten predecir la información deseada con exactitud, y se han obtenido los parámetros relevantes, que pueden ser usados por trabajadores de la industria, para predicciones futuras. Se eligió para este estudio, el arroz IRGA 424. A pesar de ser una de las variedades mayormente cultivadas en la Argentina, su comportamiento durante el secado y atemperado, no había sido reportado en la literatura al momento de comenzar esta tesis. La temperatura de transición vítrea (Tg) del arroz IRGA 424, fue satisfactoriamente determinada mediante ensayos de análisis dinámico mecánico. Tg fue calculada usando los picos de las curvas del módulo de pérdida (Tgmidpoint) y de la tangente de delta (TgEndset) en función de la temperatura. Se obtuvo un diagrama de estado, específico para esa variedad. Los valores encontrados para IRGA 424 aumentaron de 42.13 a 104.69 ° C, a medida que el contenido de humedad de las muestras disminuyó de 22.3% a 9.3% en base húmeda. Mediante estimadores estadísticos, cinco modelos matemáticos fueron evaluados para determinar su capacidad de predecir la temperatura de transición vítrea en función del contenido de agua de las muestras de arroz: Gordon y Taylor simple y linearilizada (Gordon y Taylor 1952); Fox (Fox, 1956); Jenkel y Heusch (Jenkel y Heusch, 1953) y Kwei (Kwei, 1984). El mejor ajuste se obtuvo con la ecuación de Gordon-Taylor linealizada (Gordon y Taylor, 1952). Se desarrolló un nuevo modelo matemático para predecir la temperatura de transición vítrea de IRGA 424, cuya evaluación estadística arrojó resultados satisfactorios. Se deben minimizar los gradientes de contenido de humedad dentro del grano y las diferencias de estado del material para lograr rendimientos de arroz más altos. Los valores operativos óptimos para lograr dicho objetivo son temperatura de secado igual a 40.0 ° C, tiempo de atemperado de 40 minutos y contenido de humedad al comienzo del atemperado de 18.8%. En estas condiciones, el grano alcanza un 13% de contenido de humedad en 105 minutos. Secar o atemperar arroz IRGA 424 a una temperatura de 40 °C es la mejor alternativa, ya que el cereal permanece en la región vítrea durante todo el proceso. Los parámetros del secado en capa delgada fueron calculados a partir del ajuste de siete ecuaciones de secado de la literatura: Lewis (Lewis, 1921); Page (Page, 1949); Henderson y Pabis (Henderson and Pabis, 1961); Logarithmic (Yaldiz and Ertekin, 2001); Two terms (Henderson, 1974), Midili (Midili et. al. 2002) y Logístico (Chandra and Singh, 1995). La mayor bondad de ajuste se obtuvo con el modelo de Page. Se observó que el parámetro n de este modelo no se vió influenciado por la temperatura de secado. Rice is a staple food for more than two thirds of the world’s population. It is composed of carbohydrates, proteins, vitamins and virtually no fat. Whole grain rice yields and process time could be optimized through the study of the drying process. This may result in a larger fraction of the crop entering the human food chain, enhancing the value of the crop, and resulting in a smaller fraction of broken grain, which command a lower market value.In this study, several mathematical models have been fitted to experimental data and then compared by means of a meticulous statistical analysis. In this way, equations that can be used to predict the desired information with accuracy were chosen, and relevant parameters were obtained, which can be used by industry workers for future predictions. No data of the drying behaviour of rice IRGA 424, a variety widely used in Argentina, was available before this work. Glass transition temperature (Tg) of IRGA 424 was satisfactorily determined by dynamic mechanical analysis tests. Tg was calculated using the peaks of the curves of the loss modulus (Tgmidpoint) and the tangent delta (TgEndset) as a function of temperature. A state diagram was obtained, specific for that cultivar. The values found for the rice variety IRGA 424 increased from 42.13 to 104.69 ° C, as the moisture content of the samples decreased from 22.3% to 9.3%. Using statistical estimators, five mathematical models were evaluated to determine their ability to predict the glass transition temperature as a function of the water content of the rice samples: Gordon y Taylor simple and linearilized (Gordon y Taylor 1952); Fox (Fox, 1956); Jenkel y Heusch (Jenkel y Heusch, 1953) and Kwei (Kwei, 1984). The best fit was obtained with the linearized Gordon-Taylor equation.A new mathematical model was developed to predict the glass transition temperature of IRGA 424, whose statistical evaluation produced satisfactory results. In order to preserve the quality of the rice, the gradients of moisture content within the grain and differences in the state of the material to achieve higher rice yields must be minimized. The optimal operating values to achieve this objective are drying temperature equal to 40.0 ° C, temperature time of 40 minutes and initial moisture content at the beginning of the tempering of 18.8%. Under these conditions, the total processing time needed to reach 13% moisture content is 105 minutes. Drying and tempering rice IRGA 424 at a temperature of 40 ° C is the best alternative of all studied, since the rice remains in the glassy region throughout the process. En esta Tesis se investigan y analizan tres técnicas para el desarrollo de plataformas de biosensado Lab-On-a-Chip (por sus siglas en inglés, LoC). Se presenta una técnica que permite aumentar la precisión en la cuantificación de cantidades reducidas de fluidos en aplicaciones de transporte electroosmótico y dos técnicas de detección óptica con aplicación en biosensado. Estas técnicas intervienen directamente en la automatización de procesos para la cuantificación de un elemento biológico específico o analito usando cantidades muy reducidas de fluidos. En la evolución tecnológica de las plataformas LoC se observa un constante incremento de complejidad requerida tanto en el desarrollo de sistemas de transporte microfluídico, bioreceptores y elementos transductores, como del empleo de técnicas de análisis y procesamiento de señales. En el área de transporte de especies en sistemas microfluídicos, el trabajo de Tesis está dirigido a la introducción de un modelo numérico basado en presiones de Laplace, el cual permite explicar el origen y la dinámica de gradientes de presión adversos indeseados, que pueden reducir e incluso cancelar el flujo, presentes en aplicaciones de transporte electroosmótico con mayor precisión a la alcanzada por modelos previos basados en presión hidrostática. Esto se traduce en un aumento de la precisión en el manejo de cantidades reducidas de fluidos en sistemas de transporte electroosmótico. Con respecto a las técnicas de biosensado que pueden ser implementadas en plataformas tipo LoC, en este trabajo de Tesis se explota el uso de la óptica. Se desarrollan dos abordajes bien diferenciados tanto en forma teórica como experimental: el empleo de un marco de referencia interferométrico de geometría dual basado en detección de fase usando análisis deseñales mediante el empleo de transformadas synchrosqueezing, y la aplicación de sistemas de detección basados en resonadores ópticos circulares del tipo whisperinggallery modes (por sus siglas, WGM). Ambas modalidades son discutidas en forma general utilizando experimentos controlados que se independizan de la naturaleza del elemento biológico. De esta manera los análisis presentados ponen en evidencia los pros y contras de ambas técnicas y marcan un hito de lo que se espera para su futura adaptación en el uso de plataformas tipo LoC. In this Thesis, we investigate and analyze three techniques for the development of Lab-On-a-Chip (LoC) biosensing platforms. We present a technique that allows increasing the handling precision of reduced fluid volumes in electroosmotic transport applications and two biosensing capable optical detection techniques. These techniques directly intervene in the automation of processes for the quantification of a specific biological element or analyte using minimal amounts of fluids. The technological evolution of LoC platforms shows a constant increase in the required complexity both for the development of microfluidic transport systems, bio-receptors and transducer elements, as of the use of analysis and signal processing techniques. Regarding the area of species transport in microfluidic systems, this Thesis is aimed at the introduction of a numerical model, based on Laplace pressures, which explains the origin and dynamics of undesired adverse pressure gradients, that appear in electroosmotic transport applications, which can reduce and even cancel the flow with greater precision than that achieved by previous models based on hydrostatic pressure. This renders in a precision increase in the handling of reduced amounts of fluids in electroosmotic transport systems. Regarding the biosensing techniques that can be implemented in LoC platforms, in this Thesis, we make use of optics. Two approaches are developed, differentiated both theoretically and experimentally: (1) the use of a dual-geometry interferometric reference framework based on phase detection employing signal analysis through the use of synchrosqueezing transforms, and (2) the application of detection systems based on whispering-gallery modes optical microresonators (for short, WGM). Both methods are discussed in a general way using controlled experiments independent of the biological element nature. In this way, the presented analyzes highlight the pros and cons of both techniques and mark a milestone towards its future adaptation in LoC platforms.
