En esta tesis se trabajó sobre técnicas de incremento de la sensibilidad en análisis cuantitativo utilizando herramientas ópticas. Con el foco puesto en aplicaciones biológicas y químicas, las estrategias utilizadas se pueden dividir en dos partes. Por un lado se aprovecharon las propiedades únicas de los lantánidos en la fabricación y aplicación de nanosensores con muy alta relación señal/fondo, de particular importancia en el análisis de líquidos túrbidos y medios altamente dispersivos. Con este fin se construyó un equipo para medir simultáneamente tiempo de vida y espectro de partículas luminiscentes por upvonversion (conversión ascendente) y se lo utilizó en su caracterización espectral, que tiene en cuenta sus características estacionarias pero también su dinámica resuelta en el tiempo. Por otra parte se implementó un microscopio basado en Pticografía de Fourier, una técnica que añade contraste de fase a un microscopio de transmisión tradicional y permite incrementar la resolución hasta el límite de difracción. Es de particular interés en la generación de capturas de gran tamaño con muy buena capacidad resolvente, dotando a su vez al sistema de sensibilidad de fase para la medición de espesores e índices de refracción. Se construyó un prototipo de microscopio y se trabajó en técnicas para incrementar su robustez, necesaria para el desarrollo de un instrumento de campo. Se introdujo a su vez una técnica de información estadística basada en el concepto de Información de Fischer para encontrar las zonas de máximo contenido informacional de acuerdo a la aplicación y finalmente optimizar la adquisición en tiempo y desempeño. En conjunto, ambas técnicas permiten estudiar procesos con mínima perturbación al medio observado, favoreciendo la observación de fenómenos biológicos y químicos en su contexto natural con bajo ruido y alto poder resolvente. Se han desarrollado estrategias de vinculación tecnológica para su introducción en soluciones industriales específicas tales como el sensado de efluentes y líquidos en intercambiadores de calor o el análisis petrográfico en la industria petroquímica y así demostrar su utilidad y potencialidad práctica. This thesis is centered in the implementation of optical tools to improve the sensitivity of quantitative measurements. Aiming to assess typical problems in biological and chemical applications, the strategies can be divided in two areas. On the one hand, the unique properties of lanthanides were successfully used in our advantage to construct nanosensors with a high signal/background ratio, of particular importance in the analysis of turbid or highly scattering media. With this objective in mind, a spectrometer was adapted to measure simultaneously the lifetime decay and stationary spectra of upconverting nanoparticles, allowing for the spectral characterization in both dynamical and stationary domains. On the other hand, we built a Microscope based in Fourier Ptychography allowing phase contrast imaging and resolution increase up to the limit of diffraction. This technique is perfectly suitable for large field of view applications with no loss of resolving power, with additional sensitivity to measure refractive indexes and sample heights. A laboratory prototype was constructed and used to elaborate robustness improvement techniques, a crucial need in an industrial environment. As a whole, both techniques provide a mean to study media in a minimally invasive manner, favoring the observation of biological and chemical phenomena in their natural environment with low noise and high resolving power. Technology transfer strategies were developed to introduce their application in specific industrial solutions such as effluent characterization and heat interchanging systems analysis or thin sample petrographic imaging in the petrochemical industry thus demonstrating the practical potential of the presented techniques. A partir de la necesidad de tener que incrementar la eficiencia de las plantas de generación termoeléctrica para bajar sus emisiones, se han desarrollado variantes de los aceros 9 Cr que, modificando el contenido de algunos microaleantes, buscan aumentar la resistencia a su principal carga de trabajo, el creep. La adición y optimización de los niveles de boro es uno de los ejemplos de la evolución de estos materiales. La soldabilidad de estos aceros, para el caso, es un factor crítico debido a que son equipos de construcción soldada. Es por ello que el campo de la soldadura viene acompañado de la misma tendencia que las aleaciones fundidas, tomando como primera medida la modificación de la formulación de los electrodos empleados para la soldadura por arco eléctrico de los componentes de una planta. Como sucede en la aleación de base, la optimización de los niveles de boro en los depósitos de soldaduras 9Cr con resistencia al creep mejorada (CSE9Cr) persigue reducir la velocidad de coalescencia de los carburos M23C6 ubicados principalmente en bordes de granos y sub-granos y brindar un mayor anclaje al movimiento de las dislocaciones durante la deformación. Pero no todo el control de las propiedades mecánicas de estas soldaduras se logra modificando la composición química del metal depositado, es necesario también aplicar isotermas que logren una adecuada combinación de todos los mecanismos de endurecimiento que actúan desde el principio de la deformación en la microestructura. Por consiguiente, los efectos de los parámetros del tratamiento térmico post soldadura (PWHT) aplicado, tales como las temperaturas, los tiempos de solubilizado y de revenido, tendrán marcada relevancia también en el comportamiento de la unión en servicio. Se comenzaron los estudios preliminares buscando determinar el efecto de los parámetros del PWHT sobre la evolución microestructural del metal de soldadura de un CSE9Cr que registraban incrementos en el contenido de boro respecto de un electrodo comercial. Los depósitos fueron obtenidos de dos electrodos de alambre tubular flux-cored experimental, mediante un proceso semiautomático (FCAW) bajo protección de gas Ar-20%CO2. Con dichos consumible se soldaron una serie de cupones de metal de aporte puro (AWM), elaborados según los establecido en la norma ANSI/AWS A5.29/A5.29M:2010, presentando niveles de 20 y 60 ppm de boro en su composición química. Se realizaron PWHT a distintas temperaturas y tiempos. Se extrajeron un conjunto de muestras para realizar una batería inicial de experiencias. Se contemplaron principalmente ensayos de tenacidad y dureza a temperatura ambiente y de tracción en caliente (HTT) como una alternativa a ensayos de creep, aprovechando la ventaja de obtener resultados en cortos períodos. Se aplicaron equivalencias entre los datos de HTT y de rotura al creep sobre la base de las relaciones de Norton y de Monkman-Grant, extrapolando sus resultados a tiempos más prologados por métodos paramétricos como el de Larson-Miller. Una segunda etapa, tuvo como objeto principal estudiar la evolución microestructural durante la deformación a altas temperaturas de muestras extraídas de los cupones soldados en la primera etapa y a los cuales se les aplicó los PWHT que surgieron de los resultados de los estudios preliminares. Principalmente se llevó adelante un esquema de degradación dentro de un mismo intervalo de tiempo, aplicando para el caso una fatiga termomecánica de bajo de ciclos de deformación. Mediante la difracción de rayos X (XRD) in-situ se registró la evolución de las microestructuras mientras avanzaba dicha degradación. Se correlacionaron datos de propiedades mecánicas con parámetros microestructurales obtenidos de XRD. La aplicación de PWHT de solubilizado más un doble revenido a tiempos prolongados generaron microestructuras con mayor contenido de mecanismos de refuerzo que mitigaron la degradación a alta temperatura. Con microestructuras de partida prácticamente iguales, tomando como referencia los PWHT aplicados, se observó que las muestras con 60 ppm de B comienzan a esgrimir indicios de degradación luego de haber recibido más deformación, dando cuenta de un efecto favorable respecto del contenido de B. Como evidencia de esto se observó un menor engrosamiento de partículas M23C6 y ritmo más pausado ante la pérdida de dislocaciones durante la deformación aplicada. The efforts to improve the thermal power generation plants efficiency, looking for reducing their emissions, mobilized the development of 9 Cr steels variants. Modifications in the chemical composition related to some microalloying elements to increase the resistance to creep were proposed. The addition and optimization of boron levels are examples of this evolution. Considering that many components and equipment of these plants are of welded construction, weldability is considering as a critical factor. Modifications to the formulation of the electrodes used for arc welding have been applied. Optimization of boron levels in the weld deposits of creep strength enhanced 9Cr steels (CSE9Cr) has been incorporated. From a metallurgical point of view, the reduction of the coalescence rate of M23C6 carbides at grain and subgrain boundaries, as well as the decrease of dislocation mobility during deformation, were the objectives pursued. To adjust the microstructure in order to obtain the required mechanical properties of these welds, it is also necessary apply PWHT that optimize the combination of the different strengthening mechanisms. The PWHT temperature and time will be the parameters that will define the behavior of the welded joint. In this work, initial studies showed the effect of PWHT parameters on the microstructural evolution of CSE9Cr weld metal. Two experimental flux-cored tubular wire electrodes designed to deposit 20 and 60 ppm of boron in the weld metal, were used in an FCAW process with Ar-20%CO2 shielding gas. AWM coupons were built according to the specifications of ANSI/AWS A5.29/A5.29M:2010 standard. PWHT was performed at different temperatures and times. A set of samples was extracted to carry out initial experiments. Toughness and hardness tests were conducted at room temperature, and HTT tests were used as an alternative to creep tests. Equivalences between HTT and creep rupture data were applied based on the Norton and Monkman-Grant relationships. The results of these tests were extrapolated using the parametrization proposed by LarsonMiller. The evolution of microstructure during high-temperature deformation was the objective of the second stage of the studies. PWHT parameters were applied based on preliminary results. A degradation scheme was carried out within the same time interval. A low-cycle thermomechanical fatigue was applied to achieve the degradation of the microstructures. In-situ XRD reported the evolution of these microstructures. Mechanical property data were correlated with microstructural parameters obtained with XRD. Solubilization PWHT combined with double tempering produced microstructures with the better microstructural characteristics that mitigated high-temperature degradation. Samples with 60 ppm boron showed signs of degradation after receiving greater deformation. A low growth rate of M23C6 particles and a lower loss of dislocations were observed as evidence. Las interfaces cerebro-máquina (Brain-Machine Interface (BMI) o Brain-Computer Interface (BCI)) son sistemas que establecen una comunicación directa entre el cerebro y un dispositivo externo, como una computadora o una prótesis. En el área de las tecnologías de asistencia y la rehabilitación motora, las BMIs han surgido como una poderosa herramienta cuyo potencial puede transformar la vida de personas con discapacidades motoras. En este trabajo se abordan desafíos relacionados a la portabilidad, la autonomía y la precisión en la detección de la actividad neuronal para hacer de estas interfaces herramientas seguras y plausibles de ser utilizadas en la vida cotidiana. A lo largo del desarrollo de esta tesis, se diseñó un sistema que emplea microelectrodos profundos para registrar la actividad neuronal en la corteza motora de ratas. Sin embargo, la presencia de artefactos y ruido en las señales registradas ha sido un obstáculo en la obtención de datos confiables. Para superar esta limitación, se desarrollaron dos algoritmos innovadores. En primer lugar, se propuso una técnica de eliminación de artefactos que aprovecha la identificación y eliminación de segmentos de señal que no corresponden a actividad neuronal genuina. Este algoritmo, basado en umbrales adaptativos calculados a partir de cuantiles de la señal, ofrece una mayor flexibilidad y adaptabilidad a diversas condiciones experimentales. En segundo lugar, se desarrolló un nuevo método para estimar la mediana de un flujo de datos en tiempo real. La estimación precisa de la mediana es crucial para el cálculo de los cuantiles utilizados en la eliminación de artefactos y para la detección precisa de los picos de los potenciales de acción. A partir de la sinergia entre estos dos algoritmos se ha logrado una mayor precisión en la detección de señales neuronales, gracias a la eliminación efectiva de artefactos y a la implementación de algoritmos de detección más robustos. Además, la eficiencia computacional del algoritmo de estimación de la mediana permite un procesamiento de señales en tiempo real, lo cual es fundamental para aplicaciones prácticas. Finalmente, la mejora en la calidad de las señales se traduce en una mayor fiabilidad y robustez de los sistemas BMI, ampliando su potencial para aplicaciones en entornos clínicos. En conclusión, esta tesis presenta un conjunto de herramientas y técnicas que impulsan el desarrollo de interfaces cerebro-máquina portátiles y de alta precisión. Brain-Machine Interfaces (BMI or BCI) are systems that establish direct communication between the brain and an external device, such as a computer or a prosthesis. In the field of assistive technologies and motor rehabilitation, BMIs have emerged as a powerful tool with the potential to transform the lives of individuals with motor disabilities. This work addresses challenges related to portability, autonomy, and the precision of neuronal activity detection, aiming to make these interfaces safe and viable tools for everyday use. Throughout the development of this thesis, a system was designed that employs microelectrodes to record neuronal activity in the motor cortex of rats. However, the presence of artifacts and noise in the recorded signals has posed an obstacle to obtaining reliable data. To overcome this limitation, two innovative algorithms were developed. First, a novel artifact removal technique was proposed, leveraging the identification and elimination of signal segments that do not correspond to genuine neuronal activity. This algorithm, based on adaptive thresholds calculated from signal quantiles, offers greater flexibility and adaptability to various experimental conditions. Second, a new method was developed to estimate the median of a data stream in real time. Its hardware implementation, such as on FPGA, makes it particularly attractive for portable BMI systems. Accurate median estimation is crucial for calculating the quantiles used in artifact removal and for the precise detection of action potential peaks. The synergy between these two algorithms has led to improved accuracy in detecting neuronal signals, thanks to effective artifact removal and the implementation of more robust detection algorithms. Additionally, the computational efficiency of the median estimation algorithm enables real-time signal processing, which is essential for practical applications. Improved signal quality translates to greater reliability and robustness of BCI systems, expanding their potential for clinical applications. In conclusion, this thesis presents a set of tools and techniques that advance the development of portable and high-precision brain-machine interfaces. Se han utilizado diversos algoritmos de machine learning para la estimación de la permeabilidad equivalente en medios porosos artificiales a partir de indicadores de conectividad que surgen de la configuración geométrica de los mencionados medios. Se ha elegido abordar el problema desde una perspectiva del análisis masivo de datos que ha alcanzado un impulso notable en el último tiempo.
