Durante las próximas décadas, las ciudades costeras y los deltas estarán sometidos tanto a los impactos del cambio climático como a los incrementos poblacionales. Los tomadores de decisión deberán afrontar un contexto de incertidumbre, generando estrategias robustas y flexibles para adaptarse a cualquier situación futura. El caso de estudio de esta tesis es el Delta del Río Paraná, en Argentina, uno de los sistemas de humedales más grandes del mundo. Debido a las características de sus suelos y a su cercanía con el Aglomerado de Buenos Aires, este territorio enfrenta dos grandes presiones: productiva y de desarrollo urbano. Entonces, ¿es posible cambiar el paradigma actual de la región, vinculando el sector productivo con los desarrollos urbanos, garantizando un proceso participativo que prepare a los habitantes para los desafíos climáticos venideros? En respuesta a este interrogante, esta tesis propone implementar la Planificación Adaptativa de Deltas mediante el uso de Rutas Dinámicas de Políticas Adaptativas (RDPA) para desarrollar un Sistema Soporte de Decisión (SSD). Para ello, se realizaron entrevistas con expertos y cuatro talleres en los que se probaron dos enfoques metodológicos, en la búsqueda del formato que mejor se adaptase al contexto local. Estas actividades participativas permitieron conocer las preferencias de los actores para su visión del Delta que desean para el 2050, seleccionar las acciones para lograr dichos objetivos e identificar los inconvenientes que surgen de un proceso de planificación participativo. Con la información recabada en dichas actividades, se delinearon las premisas para la construcción de una herramienta SSD. Posteriormente, se desarrolló un tablero que permitiría a los decisores seleccionar, desde las acciones y sus indicadores de interés, hasta los parámetros para materializar su visión para el Delta 2050, a través de un mapa de rutas dinámicas. Finalmente, este estudio propone una serie de recomendaciones tanto en la ejecución de los talleres como en la metodología para adaptarla a la realidad local. Con este objetivo, se propuso adecuar la metodología para sistemas con condiciones de borde dinámicas, desdoblarla en dos etapas para adaptarla a sistemas con situaciones iniciales deficientes y modificarla para que pueda ajustarse a casos con múltiples decisores. Over the next few decades, coastal cities and deltas will be subject to both impacts of climate change and population growth. Decision makers will have to face a context of uncertainty, generating robust and flexible strategies to adapt for these unforeseen futures. This thesis’ case study is the Paraná River Delta, in Argentina, one of the largest wetland systems in the world. Due to the characteristics of its soils and its proximity to the Buenos Aires Agglomeration, this territory faces two major pressures: productive and urban development. So, is it possible to change the region’s current paradigm, linking the productive sector with urban developments, while guaranteeing a participatory process that prepares the inhabitants for the upcoming climate challenges? In response to this question, this thesis proposes to implement Adaptive Delta Planning using Dynamic Adaptive Policy Pathways (DAPP) to develop a Decision Support System (DSS). To accomplish this, interviews with experts and four workshops were held, in which two methodological approaches were tested searching the format that best suited the local context. These participatory activities allowed the acknowledgment of stakeholders’ preferences for their Delta 2050 vision, the selection of actions to achieve these objectives and the identification of drawbacks that arise from a participatory planning process. With the information collected in these activities, the bases for the construction of an SSD tool were outlined. Subsequently, a dashboard that would allow decision-makers to select actions, indicators and parameters that materialize their vision for the Delta 2050 by means of a dynamic route map, was developed. Finally, this study proposes a series of recommendations, both in the execution of the workshops and in the methodology itself, in order to adequate it to the local context. With these objectives, it was proposed to adapt the methodology for systems with dynamic edge conditions, to divide it into two stages to adjust it to systems with poor initial circumstances and to modify it so that it can be fine-tuned to cases with multiple decision-makers. Hoy en día, el desarrollo de los alimentos que promuevan la salud y el bienestar es una de las prioridades de la industria alimentaria. Existe una gran variedad de compuestos con determinada actividad funcional, que se podrían utilizar como ingredientes alimentarios. De todos ellos, los prebióticos son los que han provocado un gran interés en los últimos años, probablemente debido a la incidencia de patologías relacionadas con la función gastrointestinal. Los compuestos prebióticos son considerados fibra alimentaria pero, además, estimulan el crecimiento selectivo de determinadas especies beneficiosas (bifidobacterias, lactobacilos, etc.) de la microbiota intestinal. Teniendo en cuenta que el consumo per cápita de fibra alimentaria es bajo, una buena estrategia para incrementar la ingesta diaria de fibra consistiría en incorporar compuestos prebióticos en alimentos que sean de fácil consumo y convenientes para el consumidor actual. En este aspecto, un jugo de fruta sin pasteurizar podría ser un sustrato adecuado para el enriquecimiento con prebióticos ya que, este tipo de alimento ha ganado popularidad entre los consumidores que eligen una alimentación más saludable y libre de conservantes sintéticos. De esta manera, se obtendría un producto innovador con sus propiedades nutricionales y sensoriales mejoradas, de fácil de consumo y conveniente para aquellas personas que desean mejorar la calidad nutricional de su dieta. Por lo tanto, la presente Tesis se llevó a cabo empleando jugo de frutilla (Fragaria x annanasa) como vehículo de prebióticos. La selección de la fruta se debe al significativo aporte nutricional con atributos organolépticos atractivos, y particularmente, porque es uno de los cultivos predominantes del cinturón frutihortícola del partido de Gral. Pueyrredón (Buenos Aires, Argentina). Este producto enriquecido con prebióticos requiere del adecuado diseño de estrategias de preservación que garanticen la inactivación de microrganismos deteriorantes y patógenos, sin afectar la calidad sensorial y nutricional del producto. En este contexto, el principal objetivo de la presente Tesis Doctoral fue analizar los efectos del enriquecimiento de un producto formulado a base de jugo de frutas, con el agregado de compuestos prebióticos destinado a brindar al consumidor un alimento funcional nutritivo, sensorialmente agradable, seguro y con una vida útil prolongada. En primer lugar, se investigaron los efectos del enriquecimiento con inulina y oligofructosa sobre las propiedades físico-químicas, antioxidantes, nutricionales y sensoriales de jugo de frutilla (Capítulo 1, sección 1). Luego de comprobar que era posible el enriquecimiento del producto con los compuestos prebióticos ya que no se vieron afectados los parámetros de calidad del jugo de frutilla; en una siguiente instancia, se evaluaron diferentes alternativas de preservación para prologar la vida útil del producto. En la sección 2 del Capítulo 1, se analizó el impacto de la aplicación de agentes naturales, tales como, vainillina, geraniol y resveratrol sobre la calidad microbiológica, sensorial y nutricional, así como también, sobre los parámetros físico-químicos e indicadores de capacidad antioxidante de jugo de frutilla enriquecido con inulina y oligofructosa durante el almacenamiento refrigerado. Los tratamientos con vainillina y geraniol resultaron altamente efectivos para inhibir el crecimiento de la microflora nativa de los jugos de frutilla enriquecidos con prebióticos, aunque repercutieron negativamente sobre algunos atributos sensoriales. En cambio, la aplicación de resveratrol no logró disminuir los recuentos de la microflora nativa de los jugos, pero incrementó significativamente el contenido de polifenoles totales y la capacidad antioxidante. Por otro lado, en la sección 3 del Capítulo 1, se investigó el efecto de la aplicación de tratamientos de ultrasonido sobre la evolución de los parámetros de calidad de jugo de frutilla enriquecido con inulina y oligofructosa durante el almacenamiento refrigerado. El procesamiento con ultrasonido permitió controlar la microflora de los jugos enriquecidos con prebióticos a lo largo del almacenamiento refrigerado sin introducir cambios sobre sus atributos sensoriales. De esta manera, en el Capítulo 2 se propuso combinar dos técnicas de preservación (biopreservante y ultrasonido) con dos compuestos prebióticos (inulina y oligofructosa), con el objetivo de obtener una mayor inhibición de la microflora nativa del producto, aplicando menores intensidades de ambas barreras y minimizando el impacto negativo sobre las propiedades sensoriales y nutricionales de los jugos de frutilla. Para ello, se estudiaron dos combinaciones: inulina: oligofructosa + ultrasonido + vainillina (combinación n°1) y, por otro lado, inulina: oligofructosa + ultrasonido + geraniol (combinación n° 2). Cada uno de los factores se evaluó en tres niveles diferentes para cada combinación. Se aplicó la metodología de superficie de respuesta utilizando un diseño de Box-Benhken para hallar los niveles de cada variable que optimizaran los parámetros de calidad microbiológica, nutricional y sensorial del jugo de frutilla a los 14 días de almacenamiento refrigerado. Se utilizó la función “Deseabilidad” para optimizar simultáneamente todos los parámetros. De esta manera, se halló un tratamiento óptimo para cada combinación estudiada: Óptimo_1: proporción de inulina: oligofructosa de 5:3, 7,5 min de ultrasonido (20 °C, 40 kHz, 180 W) y 1,25 mg/mL de vainillina; Óptimo_2: proporción de inulina: oligofructosa de 5:3 y 0,23 μL/mL de geraniol. Esta herramienta estadística logró alcanzar una solución de compromiso entre las diferentes respuestas bajo estudio y permitió hallar dos tratamientos óptimos, seguros desde el punto de vista microbiológico, con mayor aporte nutricional y sensorialmente agradables durante 14 días de almacenamiento. Posteriormente, se evaluó el efecto de los tratamientos óptimos hallados sobre los parámetros de calidad global de jugo de frutilla y sobre la supervivencia de Escherichia coli O157:H7 y Listeria innocua inoculados en el jugo de frutilla (simulando una eventual contaminación), durante 14 días de almacenamiento refrigerado. Una vez optimizadas las condiciones de enriquecimiento y preservación de los jugos de frutilla, se abordó un estudio sobre la estabilidad de los compuestos prebióticos durante el almacenamiento refrigerado, usando cromatografía líquida de alta resolución (HPLC, por su sigla en inglés: High Performance Liquid Cromatography) (Capítulo 3). Además, se analizó la relación entre el consumo de los azúcares predominantes y la evolución de la flora deteriorante (hongos y levaduras) en los jugos tratados bajo las condiciones óptimas. En un paso posterior, se aplicó una estrategia de análisis alternativa a HPLC, combinando espectroscopía de infrarrojo por transformadas de Fourier (FTIR, por su sigla en inglés: Fourier Transform Infrared Spectroscopy) con análisis multivariante para comprender los cambios estructurales en la composición de azúcares y compuestos prebióticos de las muestras elaboradas con tratamientos óptimos durante el almacenamiento refrigerado. El análisis de componentes principales permitió encontrar similitudes y diferencias entre las muestras almacenadas a distintos tiempos de almacenamiento en función del contenido de azúcares solubles, compuestos prebióticos y los polisacáridos principales de la pared celular. Luego, se calibraron modelos de regresión de mínimos cuadrados parciales que se utilizaron para determinar las concentraciones de glucosa, sacarosa, fructosa y los distintos fructo-oligosacáridos que están presentes en las muestras de jugo. La espectroscopía FTIR asociada al análisis multivariante, permitió la determinación de diferentes carbohidratos presentes en jugos de frutilla en una sola medición y sin necesidad de utilizar patrones de grado analítico. Esta simplicidad experimental avalaría el uso de FTIR en la línea de producción de jugos de frutilla, contribuyendo a ahorrar tiempo en la determinación de este tipo de compuestos, de una manera amigable con el medio ambiente. Como se comentó anteriormente, los jugos de frutilla representan una fuente valiosa de compuestos antioxidantes con potenciales beneficios para la salud. Para ejercer las propiedades biológicas en el cuerpo humano, los polifenoles deben hallarse en los tejidos en concentraciones suficientes para evidenciar su efecto. Por lo tanto, resulta interesante conocer la cantidad de los compuestos con capacidad antioxidante presente en los jugos con tratamientos óptimos que quedan disponibles para su absorción intestinal. En este sentido, se investigó la estabilidad de los compuestos polifenoles y flavonoides totales, del ácido ascórbico y la capacidad antioxidante frente a la digestión gastrointestinal in vitro de jugos de frutilla tratados bajo las condiciones óptimas de enriquecimiento y preservación detalladas en el Capítulo 2, simulada a días 0, 7 y 14 de almacenamiento (Capítulo 4). Los resultados obtenidos en las etapas digestivas se compararon con los contenidos iniciales en los jugos de frutilla, es decir, previo a la digestión. En líneas generales, los compuestos con capacidad antioxidante de los jugos elaborados con el tratamiento Óptimo_1 fueron más estables frente a las condiciones gastrointestinales simuladas que la muestra control, resultando en una mayor concentración de bioactivos que quedan disponibles para su absorción intestinal. En cambio, el tratamiento Óptimo_2 perjudicó la estabilidad de los compuestos bioactivos frente a la digestión gastrointestinal simulada en los distintos tiempos del almacenamiento. Esto indica que un contenido menor de compuestos antioxidantes del Óptimo_2 quedaría disponible para la absorción intestinal en comparación con el Óptimo_1 y la muestra sin tratar. El estudio de la respuesta emocional del consumidor asociada a los alimentos se ha convertido en un área importante de investigación en la industria. El creciente interés en esta área se debe al potencial de las respuestas emocionales para proporcionar información importante acerca de los productos y las diferencias entre un conjunto de productos que van más allá de las mediciones sensoriales tradicionales. En este contexto, en el Capítulo 5, se desarrolló un cuestionario visual basado en imágenes con potencial para evaluar respuestas emocionales de nuevos alimentos, como alternativa al cuestionario clásico verbal. Posteriormente, se analizaron las diferencias en la respuesta emocional del consumidor frente a una serie de nuevos productos, en este caso, jugos de frutas y hortalizas que se obtuvo usando los dos cuestionarios (visual y verbal). De esta manera, la respuesta emocional de los consumidores se evaluó en dos escenarios: compra (donde los consumidores evaluaron el producto en su envase) y consumo (donde los consumidores probaron el producto y en simultaneo observaron su envase) y se relacionó con la intención de compra. Los resultados parecen indicar que, en la situación de compra más racional, las palabras permitieron expresar mejor las diferencias en las emociones que causaron los distintos productos. En la situación de consumo, las emociones asociadas a la experiencia sensorial se diferenciaron mejor entre productos cuando se expresaron mediante imágenes. Este enfoque permitió revelar diferencias entre los jugos de frutas y hortalizas con respecto a la respuesta emocional de los consumidores y también, permitió predecir la elección de compra, proporcionando información adicional a las mediciones hedónicas tradicionales. A modo de conclusión general, el uso de herramientas matemáticas permitió encontrar las condiciones de enriquecimiento y preservación óptimas para asegurar la calidad global de jugos de frutilla durante 14 días en refrigeración. De esta manera, la presente Tesis Doctoral contribuyó con el desarrollo de jugos de frutilla funcionales, respondiendo a las exigencias del consumidor actual. Además, se demostró el potencial que tiene la espectroscopía FTIR combinada con el análisis multivariante para determinar los azúcares y compuestos prebióticos presentes en jugos de frutilla. Teniendo en cuenta que el registro de espectros no requiere reactivos químicos y toma solo un par de minutos, la espectroscopía FTIR da como resultado una metodología rápida y amigable con el medio ambiente para proporcionar información valiosa con un fuerte impacto tanto a nivel académico como a nivel industrial. La digestión gastrointestinal in vitro resultó ser un método simple y rápido para evaluar la bioaccesibilidad de los compuestos fitoquímicos presentes en los jugos de frutilla desarrollados. También, esta Tesis contribuyó a ampliar los conocimientos en referencia a las nuevas metodologías del análisis sensorial que se aplican para comprender la decisión de compra de los consumidores y que van más allá de las mediciones hedónicas tradicionales. Nowadays, the development of foods that promote health and well-being is one of the priorities of the food industry. There is a wide variety of compounds with certain functional activity, which could be used as food ingredients. Among them, prebiotics have caused a great interest in recent years, probably due to the high incidence of pathologies related to gastrointestinal function. Prebiotic compounds are considered dietary fiber but also stimulate, the selective growth of certain beneficial species (bifidobacteria, lactobacilli, etc.) of the intestinal microbiota. Taking into account that per capita consumption of dietary fiber is low, a good strategy to increase the fiber’s daily intake would be to incorporate prebiotic compounds into foods that are easy to consume and convenient for today’s consumer. In this regard, an unpasteurized fruit juice could be a suitable substrate to be enriched with prebiotics since this type of food has gained popularity among consumers who choose a healthier diet free of synthetic preservatives. In this way, an innovative product with improved nutritional and sensory properties, easy to consume and convenient for those who want to improve the nutritional quality of their diet, would be obtained. Therefore, the present Thesis was carried out using strawberry juice (Fragaria x annanasa) as a prebiotic carrier. The fruit choice was made mainly because of its significant nutritional value with attractive organoleptic attributes, but also because strawberry is one of the predominant crops in the horticultural production belt in Gral. Pueyrredón (Buenos Aires, Argentina). The prebiotic-enriched juice requires the appropriate design of preservation strategies that ensures the spoilage and pathogenic microorganisms inactivation, without affecting sensory and nutritional quality of the product. In this context, the main objective of this Doctoral Thesis was to analyze the effects of enriching a fruit-based product with prebiotic compounds to provide a nutritional and functional food, pleasant from a sensory point of view, safe and with an extended shelf-life. First, the enrichment with inulin and oligofructose on the physicochemical, antioxidant, nutritional and sensory properties of strawberry juice was investigated (Chapter 1, section 1). After verifying that enriching a strawberry juice with prebiotic compounds was possible since quality parameters of strawberry juice were not affected; in a following step, different preservation treatments to extend the product shelf-life were evaluated. In section 2 of Chapter 1, the application of natural preservatives, such as vanillin, geraniol and resveratrol were analyzed on the microbiological, sensory and nutritional quality, as well as on the physicochemical parameters and antioxidant capacity indicators of strawberry juice enriched with inulin and oligofructose during refrigerated storage. Although the application of vanillin and geraniol had a negative effect on some sensory attributes of strawberry juices enriched with prebiotics, these treatments were highly effective in inhibiting the native microflora growth. In contrast, the application of resveratrol did not reduce the native microflora counts of juices but this treatment significantly increased total polyphenol content and antioxidant capacity. On the other hand, in section 3 of Chapter 1, the application of ultrasound treatments was investigated on the quality parameters evolution of strawberry juice enriched with inulin and oligofructose during refrigerated storage. Sonication reduced the microflora growth of juices enriched with prebiotics throughout the refrigerated storage without altering their sensory attributes. Thus, in Chapter 2 the combination of two preservation techniques (biopreservative and ultrasound) with two prebiotic compounds (inulin and oligofructose) was proposed, with the aim of enhancing the antimicrobial effect, reducing each treatment intensity and minimizing the negative impact on sensory and nutritional properties of strawberry juices. With this purpose, two combinations were studied: inulin: oligofructose + ultrasound + vanillin (combination 1) and, on the other hand, inulin: oligofructose + ultrasound + geraniol (combination 2). Each factor, in each combination, was evaluated at three different levels. The response surface methodology was applied using a Box-Benhken design in order to find each variable optimal levels that optimize microbial, nutritional and sensory quality of juices after 14 days of refrigerated storage. The Desirability function was used to simultaneously optimize all parameters. In this way, an optimal treatment for each combination was found: Optimum_1: inulin: oligofructose ratio of 5: 3, 7,5 min of ultrasound (20 °C, 40 kHz, 180 W) and 1,25 mg/mL of vanillin; Optimum_2: inulin: oligofructose ratio of 5:3 and 0,23 μL/mL of geraniol. The statistical approach reached a compromise between different responses under study and allowed finding two optimal treatments, safe from the microbiological point of view, with improved nutritional and sensory properties during 14 days of storage. Then, the application of optimal treatments on quality parameters of strawberry juice and the treated juices performance against inoculated Escherichia coli O157: H7 and Listeria innocua instrawberry juice (simulating an eventual contamination), during 14 days of refrigerated storage were evaluated. Once the conditions for enrichment and preservation of strawberry juices were optimized, a study on the prebiotic compounds stability during refrigerated storage was carried out using High Performance Liquid Chromatography (HPLC) (Chapter 3). In addition, the relationship between the sugars consumption and the spoilage flora evolution (yeasts and molds) in treated juices under optimal conditions was analyzed. Then, an alternative approach was applied combining Fourier Transform Infrared Spectroscopy (FTIR) with multivariate analysis to analyze the structural changes in sugar and prebiotic compounds of samples processed with optimal treatments during refrigerated storage. The principal component analysis was useful to find similarities and differences between samples stored at different storage times which were related to changes in soluble sugars content, prebiotic compounds and the main cell wall polysaccharides. Then, different partial least square regression models (PLS) were calibrated to determine glucose, sucrose, fructose, and fructo-oligosaccharides concentrations that are present in juice samples. FTIR spectroscopy associated with multivariate analysis allowed the determination of different carbohydrates present in strawberry juices in one measurement and without using any analytical grade standards. This experimental simplicity supports the use of FTIR at the production line and contributes to save time in determining carbohydrates composition and stability, in an environmental friendly way. As it was mentioned before, strawberry juices are a valuable source of antioxidant compounds with potential health benefits. To exert the biological properties in the human body, polyphenols must be found in the tissues in sufficient concentrations. Therefore, it is interesting to know the content of antioxidant capacity compounds in juices treated under the optimal conditions of enrichment and preservation detailed in Chapter 2 that would be available for intestinal absorption. In this regard, the stability of polyphenols and total flavonoids, ascorbic acid and antioxidant capacity to in vitro gastrointestinal digestion of treated strawberry juices simulated at 0, 7 and 14 days of storage, were studied (Chapter 4). The results obtained in digestive stages were compared with the initial contents in strawberry juices (prior to digestion). In general, the antioxidant capacity compounds of juices processed with Optimum_1 treatment were more stable to simulated gastrointestinal conditions than control sample, resulting in a higher concentration of bioactives that are available for intestinal absorption. On the other hand,Optimum_2 treatment impaired the bioactive compounds stability to simulated gastrointestinal digestion in different storage times. This fact indicates that a lower antioxidant compounds content of Optimum_2 would be available for intestinal absorption compared to Optimum_1 and untreated sample. The study of consumer emotional response associated with food has become an important area of research in food industry. The growing interest in this area is due to the potential of emotional responses to provide information about a certain product or to establish differences among a set of products that go beyond traditional methods. In this context, in Chapter 5, a visual questionnaire based on images with potential to evaluate emotional responses from new food products was developed, as an alternative to the classical verbal questionnaire. Later, differences in the obtained consumer’s emotional response to a set of new products, in this case, fruit and vegetable juices using the two questionnaires (visual and verbal) were analyzed. In this way, the consumer’s emotional response and their purchase intention were evaluated in two scenarios: purchase (where consumers evaluated only the labels of the juices) and consumption (where consumers tested each juice by simultaneously analyzing its corresponding label). The results indicate that, in the rational purchasing situation, words better describe differences in emotions caused by different products. In the consumption situation, the associated emotions with sensory experience were better differentiated between products when they were expressed through images. This approach allowed to show differences between fruit and vegetable juices with respect to the consumers’ emotional response and allowed to predict the purchase choice, providing additional information to the traditional hedonic methods. As a general conclusion, the use of mathematical tools allowed to find the optimal enrichment and preservation conditions to ensure the strawberry juice overall quality during 14 days of refrigerated storage. In this way, this Doctoral Thesis contributed to the development of functional strawberry juices, responding to today’s consumer demands. In addition, the potential of FTIR spectroscopy combined with multivariate analysis to determine the sugars and prebiotic compounds present in strawberry juices, was demonstrated. Considering that spectra registration does not require chemical reagents and takes only a couple of minutes, FTIR spectroscopy results in a rapid and environmental friendly methodology to provide such valuable information with strong impact at both an academic and an industrial level. In vitro gastrointestinal digestion proved to be a simple and rapid method to evaluate the phytochemicals bioaccessibiliy of the developed strawberry juices. Besides, this Thesis contributed to broaden the knowledge in reference to new methodologies of sensory analysis applied to analyze the consumers’ purchasing decision and that go beyond traditional hedonic methods.
